人鼠肝组织嵌合体模型的制备

摘要 目的为制定人鼠嵌合HCV感染小鼠模型做初步研究。方法将人骨髓干细胞,经肝脏对KM小鼠胎鼠,每只移植1×106人骨髓干细胞,用放射免疫法对移植小鼠肝脏中的甲胎蛋白进行免疫组织化学检测,分析人肝细胞在小鼠体内生长情况。结果移植后五周内可检测到甲胎蛋白。结论将人骨髓干细胞移植小鼠肝脏内能够存活并分化成肝细胞。 Objective To establish the human mouse chimeric model of HCV. Methods The human bone marrow stem cells were transplanted into each mouse with the number of 1 × 109 via liver to fetal mouse in KM mouse. Testing the AFP in the transplanted mouse liver by immunohistochemistry to analyze the living condition of human liver cell in the mouse body. Results AFP was tested after the transplantation of months within its normal value. Conclusion The human bone marrow stem cells can live and differentiate into the liver cells after transplantation into mouse liver.
