摘要
目的:建立牛大力药材中高丽槐素的RP-HPLC含量测定方法。方法:采用Cosmosil5 C18-ms-Ⅱ色谱柱(4.6mm×250mm,5μm),柱温30℃;流动相为乙腈-0.1%醋酸水溶液(43∶57),流速1.0mL.min-1;检测波长为312nm;进样体积为20μL。结果:高丽槐素进样量与峰面积呈现良好的线性关系(r=0.9999),线性范围为0.13~2.0μg;平均加样回收率(n=6)为99.3%,RSD=1.5%。结论:该方法准确、快速、简便,重复性好,可用于牛大力药材的质量控制。
Objective:To establish an RP-HPLC method to determine the content of maackiain in the roots of Millettia speciosa Champ..Methods:A Cosmosil 5C18-ms-Ⅱ(4.6 mm×250 mm,5 μm) column was adopted,and the mobile phase consisted of acetonitrile and 0.1% acetic acid solution(43:57) at a flow rate of 1.0 mL·min-1.Detection was at 312 nm with injection volume of 20 μL.Results:The linear range was 0.13-2.0 μg,r=0.9999,and the average recovery(n=6) was 99.3% with RSD of 1.5%.Conclusion:This method is accurate,quick,simple and reproducible,which can be used for the quality control of the roots of Millettia speciosa.
