摘要
目的优化透析法除玉米须多糖中小分子杂质的工艺条件。方法苯酚-硫酸法测定多糖含量。在单因素基础上,通过3因素3水平的正交试验研究透析法除去小分子杂质的工艺,3因素分别为更换缓冲液次数、透析时间和透析温度。结果透析液与缓冲液体积比为1∶50的最佳透析条件为透析时间为7h,温度为30℃,更换缓冲液次数为3次。结论多糖纯度由原来的27.0%提高到42.35%,多糖的平均损失率9.25%。
Objective To optimize the processing conditions of removing small molecule impurities from polysaccharides by dialysis.Methods Based on single factor,the orthogonal experiments of 3 factors and 3 levels were done to remove small molecule impurities;the three factors were the number of replacement buffer,time of dialysis and dialysis temperature.Results The phenol-sulfuric acid method was used to assay polysaccharide content.The best condition of orthogonal experiment was dialysis time for 7 h,the temperature at 30℃,changing buffer frequency for 3(volume ratio of fluid and the buffer is 1∶50).Conclusion Polysaccharide content is increased from 27.0% to 42.35% and loss rate of is 9.25%.
