摘要
目的:探讨体外不同剂量电化学治疗后导致宫颈癌细胞系离子浓度改变及对其生长抑制作用的可能机制。方法:体外采用不同电压和电量组合的电化学治疗剂量处理宫颈癌细胞系,自动生化分析仪法、MTT法和活细胞计数法测定不同电化学治疗剂量处理宫颈癌细胞系前后阳、阴电极周围K+、Na+、Ca2+、Mg2+和Cl-离子的浓度、肿瘤杀伤抑制率和细胞生长曲线。结果:低电压高电量或高电压高电量组处理细胞能引起电极周围细胞Na+、Ca2+、Mg2+和Cl-离子浓度明显变化(P<0.01),而低电压低电量或高电压低电量组处理则各离子浓度无明显变化,P>0.05。各电化学剂量作用后均对肿瘤细胞产生杀伤作用。对细胞生长抑制作用强弱顺序为(5V,10C)>(10V,10C)>(10V,5C)>(5V,5C)。电化学治疗剂量(5V,10C)与(10V,10C)对宫颈癌细胞生长曲线有明显的抑制作用,P<0.01。结论:电化学治疗肿瘤细胞能引起细胞内外的Na+、Ca2+、Mg2+和Cl-离子浓度明显变化,其对肿瘤细胞杀伤抑制作用与电压电量的组合相关。
OBJECTIVE:To explore the possible mechanisms on the changes in ion concentration and growth inhibition for cervical cancer cell line with the electrochemical treatment in vitro. METHODS:The automatic biochemical analyzer,MTT and living cell count were used to detect the changes of the K+,Na+,Ca2+,Mg2+,and Cl-ion concentrations,cells' inhibition rates and growth curves of cervical cancer cells before and after electrochemical treatment with different voltage and capacity combinations in vitro. RESULTS:There were significant changes of Na+,Ca2+,Mg2+,and Cl-ion concentrations in electrodes around cells treated with the low-voltage and high power or high voltage and high power group (P0.01). The cervical cancer was inhibited by electrochemical treatment with the various doses in vitro. The order of cell growth inhibition strength was (5 V,10 C)(10 V,10 C)(10 V,5 C)(5 V,5 C). The growth curve of cervical cancer cells was significantly inhibited by electrochemical therapeutic dose (5 V,10 C) and (10 V,10 C,P0.01). CONCLUSIONS:The Na+,Ca2+,Mg2+,and Cl-ion concentrations of cervical cancer cell inside and outside will be changed markly by electrochemical treatment. The inhibitory effect on tumor cell destruction also will be associated with the combination of voltage electricity.
