新生大鼠缺氧缺血性脑损伤热休克蛋白70合成与TNF-α的关系 被引量:2

Relationship between HSP70 production and TNF-α in neonatal rats after hypoxic-ischemic brain damage

摘要 目的探讨新生大鼠缺氧缺血性脑损伤(HIBD)后HSP70合成与TNF-α的关系。方法 7日龄SD大鼠54只,随机分为空白对照组、假手术组和实验组(HIBD后2、6、12、18、24、48、72h组)共9组。用HE染色法观察脑皮质细胞形态变化,免疫组织化学法测定脑皮质HSP70阳性细胞数,放射免疫平衡法测定TNF-α含量。结果 HIBD后2h即可在皮质部位观察到HSP70阳性细胞数,HIBD后48h达到高峰,72hHSP70阳性细胞数仍比对照组高(P<0.05)。HIBD后2h,实验组大鼠脑组织出现TNF-α,12h达到高峰,然后逐渐减少,HIBD后48、72hTNF-α含量明显减少(P<0.05)。结论 HSP70和TNF-α共同参与了新生大鼠缺氧缺血性脑损伤病理生理过程,两者之间可能存在某种联系。 Objective To explore the relationship between HSP70 production and TNF-α expression in the cerebral cortex of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods Fifty-four SD rats(seven days) were randomly divided into blank control group,sham operation group,and HIBD group.Morphologic changes of cerebral cortex cells were studied with HE stain.HSP70 positive cells were counted by immunochemical methods under light microscope.Content of TNF-α was determined by radio-immunity balancing methods.Results HSP70 positive cells were found in cerebral cortex of neonatal rats after hypoxic-ischemia for 2 h,peaked after 48 h,and still significantly higher in HIBD group than in control group after 72 h(P〈0.05).TNF-α was found in cerebral cortex in HIBD group after hypoxic-ischemia for 2 h and peaked at 12 h,then decreased gradually.TNF-α decreased significantly after hypoxic-ischemia for 48 and 72 h(P〈0.05).Conclusions Both HSP70 and TNF-α participate in the pathophysiological process of hypoxic-ischemic brain damage in rats,and there might be some relation between those two factors.
