高压氧治疗对缺氧缺血性脑损伤新生大鼠脑组织p53表达的影响 被引量:1

Effect of Hyperbaric Oxygen Therapy on Expression of p53 in Cerebral Tissue of Neonatal Rats with Hypoxic-Ischemic Brain Damage

摘要 目的探讨高压氧(HBO)治疗对缺氧缺血性脑损伤(HIBD)大鼠脑组织p53表达的影响。方法将56只7日龄Wistar大鼠随机分为3组:假手术组(8只)、HIBD模型组(又分为24h、48h、72h组,每组各8只)和HBO组(24h、48h、72h组,每组各8只)。假手术组大鼠只分离其左侧颈总动脉而不结扎。HIBD模型组和HBO组大鼠结扎左侧颈总动脉2h,然后置于密闭缺氧舱(含80mL.L-1氧气、920mL.L-1氮气混合气体)2h,制备HIBD模型组。HBO组予高压氧治疗。各组分别在造模后及治疗24h、48h、72h处死大鼠,取脑。采用免疫组织化学法和免疫印迹法(Westernblot)观察各组大鼠海马区p53阳性细胞计数和p53蛋白表达情况。应用SPSS12.0软件进行数据处理。结果假手术组几乎未检测到p53阳性细胞和p53蛋白表达,与HIBD组和HBO组比较差异均有统计学意义(Pa<0.05)。HBO组各时间段海马p53阳性细胞数和p53蛋白表达均显著少于HIBD组,二组比较差异均有统计学意义(Pa<0.05)。结论 HBO治疗可降低新生大鼠HIBD后脑组织p53的表达,从而减轻其脑组织损伤。 Objective To explore the effect of hyperbaric oxygen(HBO)therapy on the expression of p53 in cerebral tissue of neonatal rats with hypoxic-ischemic brain damage(HIBD).Methods Fifty-six 7-day-old Wistar rats were randomly divided into 3 groups:sham-operated group(n=8),HBO group(n=24)and HIBD group(n=24).HBO group and HIBD group were divided into 24 hours group(n=8),48 hours group(n=8)and 72 hours group(n=8),respectively.These rats in sham-operated group were separated the left carotid arteries without occlusion.These rats in HBO group and HIBD group were prepared with the left occlusion of carotid arteries for 2 hours,following 2 hours later,breath 80 mL·L-1 oxygen with 920 mL·L-1 nitrogen mixture for 2 hours.Rats in HBO group were given HBO therapy.Eight rats were sacrificed at 24 hours,48 hours and 72 hours later,respectively.The level of p53 protein expression and the p53 positive cell counting in hippocampus were measured by immunohistochemical method and Western blot.SPSS 12.0 was used to analyze the data.Results No any p53 positive cell was found in sham-operated group,and there were significant differences compared with HBO group and HIBD group(Pa0.05).The level of p53 protein expression and p53 positive cell counting in hippocampus in different times in HBO group were significantly lower than those in HIBD group(Pa0.05).Conclusions HBO therapy can decrease the expression of p53 in hippocampus of HIBD neonatal rats,then relieve the cerebral anoxic injury.
