人催乳素单克隆抗体制备及其临床应用研究

摘要 为获得人脑垂体催乳素(prolactin,PRL)单克隆抗体,用PRL粗制品免疫了BALB/c小鼠。利用杂交瘤技术,将免疫小鼠的脾淋巴细胞与NS-1骨髓瘤细胞融合,产生杂交瘤细胞株,经过筛选,选出了2株能分泌催乳素单克隆抗体的杂交瘤细胞株PRLH6和PRLA2。利用小鼠体内诱生单克隆抗体方法获得了腹水。对杂交瘤细胞进行了染色体分析,两株杂交瘤细胞染色体数目平均分别为100条和90条;抗体效价检测均为1:10000,杂交瘤细胞产生的免疫球蛋白均为IgM。琼脂扩散试验发现该抗体与促甲状腺素(TSH)、促卵泡素(FSH)及黄体生成素(LH)无交叉反应。以纯化后的抗体作为酶联免疫吸附测定(ELISA)的包被物,用双抗体夹心法检测了20份正常人血清和30份高催乳素血症病人血清标本,发现PRL水平差异显著。该单克隆抗体可用于制备放免、酶免试剂药盒。 To abtain monoclonal antibody against human prolactin,BALB/c mice were immunized with human prolactin.By fusing mouse myeloma cell(NS-1) with spleen cell from the immunized mouse, two hybridomas cell lines (PRLH6,PRLA2) which secrete prolactin monoclonal antibody were established.The antibody was of IgM type and did not have cross reactions with TSH,FSH and LH.Serum prolactin levels in healthy controls and hyperprolactinemic patients were tested with this monoclonantibody using ELISA.The OD values were 0.2140.049 and 0.8120.325 respectively.This antibody could be used in preparation of prolactin kit.
