基因工程人催乳素(rhPRL)促进骨髓移植小鼠的造血重建

摘要 选用BACB/c鼠经致死放疗后行同基因骨髓移植(SBMT),在小鼠SBMT后腹腔注射rhPRL(10μg/次,隔日进行,共10次).结果显示,小鼠骨髓和脾脏中造血前体细胞(CFU-C和BFU-E)比例增加,外周血中WBC、RBC和PLT亦均有明显的时相提前,该剂量rhPRL无任何病理损伤和增长体重的作用,提示rhPRL在促进BMT造血重建有应用前景. Recombinant human prolactin (rhPRL) was administered to mice after syngeneic bone marrow transplantation (SBMT) to determine its effects on immunologic and hematopoietic reconstitution. BALB/c mice were given 10μg ip injections of rhPRL every other day for a total of 10 injections after SBMT. Upon analysis, mice receiving rhPRL exhibited significant increases in hematopoietic progenitor cell (CFU-C and BFU-E) contents and cellularity in both the bone marrow and spleen. Analysis of the peripheral blood indicated that administration of rhPRL resulted in significant increases in the rate of white blood cell (WBC), red blood cell (RBC) and platelet (PLT) with increased absolute numbers of lymphocytes and neutrophils. The presence of cells positive for the granulocyte marker (8C5) was also increased in the bone marrow and spleen of treated mice. No significant pathologic effects were observed and no weight gain was noted in mice receiving repeated injections of rhPRL at this dose and schedule.
