摘要
本文利用合成的降钙素基因相关肽的抗原决定簇Tyr-CGRP(27~37)制备抗CGRP抗血清,用Iodogen做氧化剂标记抗原,以免疫沉淀剂进行B与F分离,从而建立了降钙素基因相关肽的放免测定方法。方法灵敏度为6pg/mL,回收率65%~80%,批内CV776%,批间CV1138%.正常人血浆CGRP水平4357±528pg/mL,急性心梗患者血浆CGRP水平7024±623pg/mL,两者有显著差异(P<005)。体外循环病人手术期间血浆CGRP较术前有明显变化(P<005),术后24小时CGRP较术前及术中有明显升高(P<005)。
Synthetic antigenic determinant of calcitonin gene related peptide(CGRP),tyr-CGRP(2737),was used to prepare anti-CGRP antiserum.As oxidizer,iodogen was neutralized in making labeled antigen.Immunoprecipitation reagent was adopted for separating binding phase from free phase.Then,the radioimmunoassay of CGRP was established.The sensitivity was 6pg/mL,the recovery rate was 65%80%.The intra-and inter-assay coeficients of variation(CV) were 776% and 1138%.The CGRP levels in serum of healthy groups and acute myocardial infarction patients were 4357528pg/mL and 7024623pg/mL respectively.There was significant diffrence(P<005)between two group.There was difference in the serum levels of cardiopulmonary bypass pateint between operation stage and before operation stage(P<005).Twenty four hours after operation,the serum CGRP level was significantly higher than that of operation stage and before operation stage(P<005).
