降钙素基因相关肽的放射免疫测定及临床应用

摘要 本文利用合成的降钙素基因相关肽的抗原决定簇Tyr-CGRP(27~37)制备抗CGRP抗血清,用Iodogen做氧化标记抗原,以免疫沉淀剂行B与F分离,从而建立了降钙素基因相关肽的放免测定方法,方法灵敏度为6pg/ml回收率65%~80%,批内CV7.76%,批间CV11.38%,正常人血浆CGRP水平43.57±5.28pg/ml,急性心梗塞患者血浆CGRP水平70.24±6.23pg/mg。 Synthetic antigenin diterminant of Calcitonin Gene Related Peptide (CGRP), tyr-CGRP(27~37), were used to preparate anti-CGRP antiserum. As oxidizer, Iodogen was natural-ized in making labeled antigen. Immuno-pricipititing reagent was adopted for separating bindingphase from free phase. Then, the radioimmunoassay of CGRP was have been created. The sensi-tivity was 6 pg/ml. The recovery ratio was 65%~80%. The intra-and inter-assay coeficient ofvariation (CV) were 7. 76% and 11. 38%. The CGRP level in serum of healthe human group andacute myocardial infarction patient were 43. 57±5. 28 pg/ml and 70. 24±6. 23 pg/ml. There wassignificant diffrence (P < 0.05) between the two group. In cardiopulmonary bypass (CBP)pateint's serum, three was diffrence about CGRP level beteen operation stage and before operationstage. (P<0. 05) In 24 houre after operation, serum CGRP level was significant higher than bothoperation stages' and before operation stages' (P<0.05).
