摘要
利用葡萄糖和果糖在碱性条件下烯醇化反应速度不同,提出了速差动力学光度法同时测定葡萄糖和果糖的新方法。通过实验条件的选择,确定了葡萄糖和果糖与斐林试剂反应,与磷钼酸显色的动力学反应级数为零级;且反应表观速率常数分别为:0.108mg/L·min,0.427mg/L·min;测量浓度的线性范围均为:2~8mg/L;最低检测限为1.0×10-9g/L;对人工合成样品测定结果的相对标准偏差为1.20%~341%;对实际样品测定结果的标准回收率为95%~102%。
This paper presents a new method of simultaneous determination of glucose and fructose by differential reaction-rate kinetic spectrophotometry based on the difference of enolization reaction-rate of glucose and fructose in basic medium.Under optimum experiment conditions,the kinetic reaction grades of glucose and fructose reacting with Fehling reagent and phosphomolybdic acid are zero;the apparent constants of reaction-rate of glucose and fructose are 0.108 mg/L·min,0.427 mg/L·min,respectively.The linear determination ranges of glucose and fructose are 2~8 mg/L.The minmum detectability is 1.0×10 -9 g/L.The RSD% for artificial samples is 1.20%~3.41%.The recovery for real samples is 95%~102%.
