摘要
应用均匀设计法探讨了影响深山草莓花瓣愈伤组织诱导及愈伤组织再分化变异植株的主要因子和水平,并从中获得稳定的变异株系。结果显示:深山草莓花瓣愈伤组织诱导率超过65.0%以上的培养基共有11个,其中以LS+TDZ 2.06 mg.L-1+2,4-D 0.60 mg.L-1培养基的诱导效果最好,诱导率达98.0%;最佳再分化培养基为1/2LS+TDZ 2.30 mg.L-1+KT 1.38mg.L-1,田间试验发现变异率高达2.77%。再生植株生根移栽后观察筛选获得6个性状表现稳定的变异株系,再经过7年的栽培试验获得了1个性状稳定的优良变异株系。
The main factors and levels affecting callus induction and mutation rates from in vitro culture of petals of Fragaria orientalis Lozinsk.var.concolor(Kitag.) Liou et C.Y.Li were studied,and stable mutants were obtained.The results showed that eleven culture media achieved callus induction rate of more than 65.0%,among which LS+TDZ 2.06 mg·L-1 +2,4-D 0.60 mg·L-1 was the best for callus induction with a callus induction rate of 98.0% and 1/2LS+TDZ 2.30 mg·L-1+KT 1.38 mg·L-1 was the best for callus re-differentiation with a mutation rate of 2.77% following field cultivation.Six stable mutant lines were obtained through field cultivation of tissue culture plants and one stable and superior mutant was obtained following 7-years of field cultivation.
