摘要
目的:探讨凋亡在视神经撞击和挤压损伤导致视网膜神经节细胞死亡中的作用。方法:应用活细胞悬液荧光染色法和流式细胞仪 P I染色法检测损伤后视网膜神经元凋亡的形态和数量。结果:荧光染色法见视野内有散在细胞出现核染色质凝聚现象( 不规则核块) ,流式细胞仪 D N A 直方图见正常对照组有直而高的 G0/ G1 峰和低平的 G2 峰,在 G0/ G1 之前有一细胞群体,大约占细胞总数的163 % ,损伤组在 G0/ G1 之前有一低小的亚二倍体峰,大约占全部细胞的483 % 。结论:视神经不完全损伤后存在视网膜神经元的凋亡,这一结论可为视神经外伤的临床治疗提供实验依据。
Aim:To investigate the effect of apoptosis in the death of retinal ganglion cells after the optic nerve striking and extruding injuries. Methods:①Suspended retinal cells were made up with Hoechst 33 342 and observed under microscopy. ②Retinal cells were made up PI and analyzed by FCM. Results:There were a few condensation of nucleus chromatin(irregular nucleus) in injuries group (apoptosis cells). But there was no same phenomenon in normal group. Histogram of FCM analysis: there was a small apoptotic peak in injuries group, which are about 4.83% of all retinal cells. But in normal group there were only 1.63% before G 0/G 1 peak and no apoptotic peak could been seen. Conclusion:It demonstrated the apoptosis was one of the mechanisms of retina cell death after incomplete striking and extruding injuries of optic nerve. It could provide experimental basis of medicated treatment in traumatic optic neuropathy.
