瘦素对低温导致的大鼠胃黏膜应激性溃疡的预防作用研究 被引量:1

摘要 目的明确瘦素(Leptin)干预对胃黏膜中过氧化物和凋亡相关因子的影响,探讨瘦素对持续冷刺激导致的大鼠胃溃疡的作用。方法将腹腔内注射瘦素的大鼠置于4℃的环境下进行冷刺激,4h后取胃组织采用硫代巴比妥酸比色法(TBA法)检测胃黏膜中脂质过氧化主要产物丙二醛(MDA)的含量,双硫对硝基苯甲酸比色法测定谷胱甘肽(GSH)水平,通过免疫组织化学的方法检测Caspase-3的表达。结果与对照组相比,持续冷刺激组的胃MDA水平明显提高(P<0.01),GSH水平降低(P<0.01),Caspase-3表达增加(P<0.01)。Leptin干预阻止了冷刺激组动物MDA水平的提高(P<0.01)和胃黏膜中GSH含量的降低(P<0.01),Caspase-3表达也明显减少(P<0.01)。结论持续的冷刺激通过激活氧化剂降低了胃组织的抗氧化能力,通过提高Caspase免疫活性诱导凋亡。Leptin的存在逆转了这些机制并抑制凋亡,对胃黏膜具有保护作用。 【Objective】To identify effect of leptin intervention to perhydride and apoptosis correlation factor in gastric mucos membrance and investigate effect of leptin to rat gastrelcosis induced by cold-restraint stress.【Methods】Rats pretreated with intraperitoneal injection of leptin were restrained at 4°C environment confront cold stress.After 4 hours,tissues of stomach from the rats were taken.The TBA method was used for detection of malonaldehyde(MDA) level.DTNB colorimetric method was used for detection of glutathione(GSH) levels.Expression of caspases was investigated by immunohistochemistry.【Results】While the stomach MDA level of the cold-restraint stress group was increased significantly(P 0.01),the level of GSH was decreased(P 0.01),Express of caspases was increased(P 0.01) compared with the control group.Leptin administration prevented the increase in the MDA level(P 0.01) and the decrease in the GSH content(P 0.01) of the gastric mucosa in animals subjected to stress.Expression of caspases was decreased significantly(P 0.01) too.【Conclusion】Cold-restraint stress decreases the antioxidant capacity of stomach tissue while activating oxidants,and induces apoptosis by an increase in caspase immunoreactivity.The presence of leptin reverses these mechanisms and suppresses the apoptosis,which has protection to gastric mucos membrance.
