摘要
将人免疫球蛋白G(hIgG)抗体固定在多壁碳纳米管修饰的玻碳电极表面,制备了一种电化学发光(ECL)免疫传感器。以hIgG抗体标记的联吡啶钌脂质体为标记物,采用三明治型检测方式,成功建立了hIgG的ECL免疫检测技术。电化学发光强度与hIgG的浓度在0.01~0.8μg/L范围内呈良好的线性关系;线性回归方程为y=618.7x(μg/L)-23.9(n=6,r=0.995);检出限为0.004μg/L。用于人血清中hIgG的检测,结果令人满意。
A simple and sensitive electrochemiluminescence(ECL) immunosensor was fabricated by immobilizing goat-anti-human immunoglobulin G(hIgG) antibody on a multi-walled carbon nanotubes(MWCNTs) modified glassy carbon electrode.The hIgG antigen and antibody-tagged liposome can be sequentially bound on the immunosensor through antigen-antibody specific interaction,forming a sandwich-type immunocomplex.The ECL intensity of the immunosensor increased linearly with the hIgG concentration from 0.01 to 0.8 μg/L with a detection limit of 0.004 μg/L(S/N = 3).The immunosensor mentioned above was successfully applied for the detection of hIgG in human serum.
