摘要
目的探讨养心通脉有效部位方(active principle region of Yangxing Tongmai Formula,apr-YTF)促进急性心肌梗塞大鼠骨髓间充质干细胞(Mesenchymal stem cells,MSCs)定向归巢及其对心功能和病理的影响。方法将造模成功的大鼠32只,随机分为apr-YTF组、香丹注射液组、重组人粒细胞集落刺激因子(rhG-CSF)组、模型对照组,每组8只,均于造模3h后给予相应药物,连续5d;均于造模后第2天开始腹腔注射5-溴脱氧尿嘧啶核苷(Brdu),连续4d。用PcLab生物医学信号采集处理系统检测记录心功能,masson's三色染色检测梗塞心肌病理,Image-Pro Plus6.0病理图象分析系统测量梗塞区瘢痕面积百分比,免疫组化的方法检测梗塞心肌边缘区的CD34+染色阳性细胞数、Brdu与肌钙蛋白I(cTn I)双染阳性细胞数。结果 rhG-CSF组和apr-YTF组的心功能4个指标较模型组均有显著改善(P<0.05或P<0.01);两组的心肌梗塞边缘区瘢痕组织内可见新生的心肌细胞和血管,恢复的趋势较为明显;两组的心室肌瘢痕面积百分比较模型组和香丹组显著减小(P<0.01);两组大鼠梗塞心肌边缘区CD34+染色阳性的细胞数、Brdu与cTn I双染阳性的细胞数较模型组显著增加(P<0.01),apr-YTF组较rhG-CSF组显著增加(P<0.01)。结论 pr-YTF能促进MSCs归巢并因此而促进心肌细胞和心功能恢复。
Objective To explore the effect of apr-YTF(active principle region of Yangxing Tongmai Formula)on cardiac function and pathology of MSCs homing to infarcted myocardium in rats.Methods32 SD male rats with acute myocardial infarction were randomly divided into apr-YTF group,Xiangdan injection group,recombinant human granulocyte colony stimulating factor(rhG-CSF)group and model control group 8 in each group.After 3 h,myocardial were given corresponding drug for 5 d.5-bromo-deoxyuridine(Brdu)was intrapthtoneally injected on day 2 after model establishment for 4 days.Cardiac function with PcLab biomedical signal acquisition and processing system,myocardial infarction pathology was detected by masson's three-color,Image-Pro Plus 6.0 image analysis system was used to determine infarct scar area percentage.Positive cells,Brdu and cardiac troponin I(cTn I)double staining positive cells were determined by immunohistochemistry.Results Four indicators of cardiac function in rhG-CSF group and apr-YTF group were significantly improved compared with the model group(P0.05 or P0.01);myocardial infarction scar tissue was seen in the marginal zone of the myocardial cells and vascular newborn.Percentage of ventricular muscle scar area was significantly reduced compared with model group.The two groups of rats with myocardial infarct marginal zone stained positive and Xiangdan injection group(P0.01)CD34+ cells,Brdu and cTn I double staining positive cells increased significantly compared with model group(P0.01).Apr-YTF group were significantly increased compared with rhG-CSF group(P0.01).ConclusionApr-YTF can promote the homing of MSCs,and thus contribute to cardiac function recovery.
