摘要
[目的]建立彩叶草快速繁殖体系。[方法]以绯巫彩叶草的茎段为外植体,对彩叶草增殖、生根及组培苗移栽的最适培养基及生长情况进行研究。[结果]绯巫彩叶草芽苗增殖的最适培养基为MS+BA1.50mg/L+NAA0.05mg/L+水解酪蛋白0.40g/L+蔗糖30.0g/L,植株生长良好,成活率达100.0%,增殖倍数达8.65;生根培养适宜的培养基为1/2MS+NAA0.10~0.50mg/L,生根率达100.0%,平均不定根根数达12.95~17.88条;生根培养基中用白糖代替蔗糖作碳源的外植体生根率达100.00%,根多而长,植株生长良好;试管苗移栽到腐叶土中成活率为92.86%。[结论]该研究结果可为建立彩叶草大规模工厂化育苗体系提供依据。
[Objective]The aim was to establish a rapid propagation system of Coleus blumei.[Method]Stem of Coleus blumei 'Feiwu' were used as explants,the optimum media and growth situations of the proliferation,rooting culture,and transplantation of Coleus blumei 'Feiwu' were studied.[Result]The optimum medium for shoots proliferation was MS+1.50 mg/L BA+0.05 mg/L NAA+0.40 g/L casein hydrolysate+30.00 g/L sucrose,the survival rate was 100.00%,multiplication coefficient was 8.65 and the shoots grow well.The suitable rooting medium was 1/2MS+ 0.10-0.50 mg/L NAA,the rooting rate was up to 100% and each shoot could produce 13-17 roots.The white sugar was used as carbon source instead of sucrose could make shoots produce more and long roots with the rooting rate was 100.00%,the plantlet survival rate was 92.86% when the plantlets were transplanted into rotten leaf soil.[Conclusion]This study could provide theoretical and technical basis for establishing a large industrial seeding system of Coleus blumei.
