摘要
实验从骨内血流动力学和血液流变学角度探讨活血止痛汤治疗早期骨性关节炎的机理。通过手术结扎股静脉的方法造成兔右膝关节骨性关节炎的模型。造模5周后,使用活血止痛汤进行治疗。用药5周后进行检测,结果表明,活血止痛汤能显著降低骨内高压,降低骨髓血的全血粘度、血浆粘度、红细胞压积和血浆纤维蛋白原含量,和正常组相比,无显著性差别(P>0.05);和造模组相比,有高度显著性差别(P<0.01)。光镜和电镜观察发现,中药组的关节软骨和正常组相比无明显退变,而造模组的关节软骨则出现明显退行性改变。实验结果表明,活血止痛汤能通过活血化瘀,改善骨内血流动力学和血液流变学状态,从而达到保护关节软骨。
The mechanism of “HUOXUE ZHITONG TANG” (HZT, Decoction for Activating Blood Circulation and Removing Pains) treating early osteoarthritis was explored through experiments of hemodynamics and blood rheology. Osteoarthritis models were made in the right rabbit knee by ligating femoral vein. Five weeks after modelling, the models were treated by HZT. The tested results 5 weeks after the administration showed that HZT could significantly reduce intraosseous pressure, whole blood and plasma viscosity, hematocrit, and plasma fibrinogen levels of the intramedullary blood. The treated group had a significant difference from the modelled group (P < 001) but no remarkable difference from the normal group (P > 0.05). The optical and electronic microscopy revealed that the articular cartilage of the treated group had no marked retrograde change compared with the normal group, while significant retrograde changes appeared in the articular cartilage of the modelled group. The experimental results suggested that HZT functioned to protect articular cartilage, prevent and treat osteoarthritis through activating blood circulation, eliminating blood stasis, and improving intraosseous hemodynamics and blood rheology.
