摘要
目的:测定葛根芩连汤煎剂(T)和甘草单煎剂(S)中甘草次酸(Glycyrrhetinic acid,GA)家犬血清中的含量,分析煎煮方式对其体内药动学的影响。方法:建立测定家犬血清中甘草次酸的高效液相色谱法,通过测定不同时刻家犬体内甘草次酸浓度,计算药动学参数。结果:家犬灌服T和S后甘草次酸在家犬体内的Cmax分别(244.8±56.1)和(986.5±116.9)ng/mL;tmax分别为(9.7±1.5)和(10.3±1.5)h;AUC0~∞分别为(4766±963)和(14483±2410)ng/(h·mL)。结论:煎煮方式对甘草次酸家犬体内药动学有显著影响,在葛根芩连汤中甘草次酸清除半衰期明显延长。
AIM: To investigate the effect of extraction on pharmacokinetics of glycyrrhetinic acid (GA) by determining GA concentration of serum after administration of Gegen Qinlian Decoction (T) and GlycTrrhiza uralensis decoction (S). METHODS : HPLC method was established to determine serum concentration of GA and plaarmacokinetic parameters were calculated. RESULTS : The parameters in dogs after administration of T and S were as follows: Cmax was (244.8 ±56.1) and (986.5 v116.9) ng/mL; tmax was (9.7 ± 1.5) and (10.3 ± 1.5) h; and AUC0-∞ was (4 766 ± 963 ) and (14 483 ± 2 410) ng,/(h · mL), respectively. CONCLUSION: Extraction has significant effect on pharmacokinetic parameters, the removal half-life of GA in Gegen Qinlian Decoction is markedly extended.
