摘要
目的:制备柚皮苷亲水凝胶骨架片,并对其释药影响因素及释放模型进行探讨。方法:采取羟丙甲基纤维素(HPMC)为骨架材料,以湿法制粒压片法制备了柚皮苷亲水凝胶骨架片。考察了辅料(PEG类型、乳糖、蔗糖、甘露醇),HPMC分子量及含量对药物释放的影响,并采取零级、一级、Higuchi及Ritger-pappas模型,对药物的释放进行了拟合。结果:辅料对骨架片中药物释放的影响为:PEG4000>PEG400>PEG10000;填充剂为甘露醇的骨架片释放速度要快于含乳糖和蔗糖的骨架片;HPMC分子量及含量越大,药物释放越慢;药物的释放符合零级释放,药物的释放以溶蚀为主。结论:制备的柚皮苷亲水凝胶骨架片能持续缓慢地释放药物。
AIM: To prepare naringin hydrogel matrix tablet and investigate the factors that may influence drug release. METHODS : Wet method was used to prepare the sustained hydrogel matrix tablet of naringin with HPMC as the matrix material, the adjuvant (PEG of different molecules, lactose, sucrose and mannital) that may influence the drug release was studied as well as the influence of the molecular weight and amount of HPMC in the tablet. Zero-order, First-order, Higuchi equation and Ritger-pappas equation were used for the model fitting of drug release. RESULTS: The influence of the adjuvant on drug release was following PEG-4000 〉 PEG-400 〉 PEG- 10000 and mannitol had a faster drug release rate than lactose and sucrose. It had also been proved that the larger molecular weight and greater amount of HPMC, the slower release of naringin from the hydrogel matrix tablet. The drug release was in good coincidence with Zero-order in an erosion manner. CONCLUSION: The prepared naring- in hydrogel matrix tablet could release its drug in a slow and continuous manner.
