摘要
采用实时荧光PCR方法鉴别含麸质的谷类成分,选择大麦Hordein管家基因、小麦Gliadin管家基因、黑麦Sec1管家基因和燕麦Avenin管家基因作为物种鉴定的特异性标记基因,设计适合于Taqman探针实时荧光PCR扩增的引物和探针,从而达到鉴别检测食品中含麸质的谷类大麦、小麦、黑麦、燕麦成分的目的.特异性实验结果表明,分别采用大麦、小麦、黑麦、燕麦的引物和探针进行实时PCR扩增时,经检测25种样品,只有相对应的阳性物种样品产生荧光信号,其余非该物种样品均不产生荧光信号,表现出了很高的物种鉴定特异性.以大麦为例的灵敏性实验结果表明,原料样品检测灵敏性可达到0.01%,加工后的食品则可达到0.05%,符合食品成分痕量检测要求.
This paper has established a real-time PCR method which allowed detection of gluten-containing cereals in food.Primers and probes were designed on the basis of housekeeping gene of Hordein,Gliadin,Secl and Avenin.The specificity of this method was confirmed by 25 kinds of samples.The experiment results show that only positive samples produced fluorescent signal.After using primers and probe of Hordein,Gliadin,Secl and Avenin seperately.Take Hordein for example,sensitivities result shows that 0.01% of Hordein for raw material and 0.05% of Hordein for food could be detected,which meet the need of trace detection of ingredients in food.
