北冬虫夏草深层发酵高产虫草素工艺的优化

Optimization of submerged fermentation process of Cordyceps militaris to produce a high cordycepin content

摘要 对23个北冬虫夏草菌株发酵液中虫草素的含量进行了测定,发现发酵液中虫草素含量最高的菌株为21号;对该菌株的发酵条件进行优化,获得最适培养基配方为:蔗糖40g/L,蛋白胨15g/L,MgSO4·7H2O0.5g/L,KH2PO40.5g/L,K2HPO4·3H2O 0.5g/L,pH自然;在此培养基中添加0.5g/L腺嘌呤进行培养,可使发酵液中虫草素含量提高约30%,同时腺嘌呤的添加可以加快虫草素的合成。培养方式的考察结果表明:纯静置培养有利于虫草素的产生和积累,静置培养28d,所得发酵液中虫草素含量达2.05g/L,比优化前提高了2倍。 The fermented broths of 23 kinds of Cordyceps militaris strains were compared in terms of cordycepin content and it was found that Strain 21's fermented broth was the highest in cordycepin content. Optimization of the fermentation conditions for Strain 21 showed that the optimal medium was 40 g/L sucrose, 15 g/L peptone,0.5 g/L MgSO4-7H2O,0.5 g/L KH2PO4 and 0.5 g/L K2HPO4- 3H20,and its pH value was natural. Culture with this medium to which 0.5 g/L adenine was added could make the cordycepin content of the fermented broth increase by 30%. It was proved that the added adenine could accelerate the synthesis of cordycepin. The investigation of the culture conditions showed that static culture was beneficial to the synthesis and accumulation of cordycepin, and 28 d static culture could gain 2.05 g/L cordycepin in the fermented broth which was two times higher than that before optimization.
