摘要
用化学合成法,我们设计、合成了一个以植物偏爱的遗传密码子编码的天蚕抗菌肽B基因。通过把该基因与M13mp19噬菌体载体重组、克隆;转化大肠杆菌JM103,杂交检测和序列测定,得到二个与设计一致的克隆。
The cecropin B is one sort of peptides existing in the body of Hyalophra ceceropin. It can kill bacterial. We have attempted to transform such a gene to plants. A 153 bp DNA encoding cecropin B and containing appreciate restriction enzyme sites for plasmid insertion, stop codon at 3' end was chemicall synthesized, The synthesis involved the preparation of 10 oligonucleotides and onz. matic ligation. After a ligation of s nthetic gene to phage vector M13 mp19 and transformation to JM103 strain, the clones containing the anticipated gene sequence was obtained by hybridiztion. The accuracy of sequence was confirmed by DNA sequencing.
