摘要
目的探讨黄芪多糖(APS)对糖尿病(DM)大鼠尿量、肾脏水通道蛋白2(AQP-2)表达的影响。方法制备大鼠模型,并分别给予不同剂量的APS腹腔注射,6周后检测24h尿量,用免疫荧光和RT-PCR法观察AQP-2的表达。结果 DM组大鼠尿量明显增多,APS可增加DM大鼠尿量;DM大鼠肾脏髓质AQP-2表达明显增强,APS降低其高表达。结论 APS改善DM水盐代谢的机制可能与降低肾脏AQP-2表达有关。
Objective To investigate the effects of APS on the urinary volume and the expression of AQP-2 in the kidney of diabetic rats. Methods The DM rats were given APS with different dosage. Urinary volume were measured after 6 weeks of treatment. The protein expression and location of AQP-2 were observed by immunofluorescencestain. Moreover, the RT-PCR technique was used to determine the levels of AQP-2 mRNA expression in kidney medulla. Results Diabetic rats showed a significant increase in urinary volume compared with control rats. APS treatment could obviously increase the urinary volume;the expression of AQP-2 was markedly increased in DM versus contol group, and the APS could inhibit the expression of AQP-2. Conclusions APS could down-regulate the expression of AQP-2,which may be its mechanism for regulating fluid metabolism.
