17-β雌二醇对西伯利亚鲟卵黄蛋白原的诱导及检测 被引量:2

Induction and determination of Vitellogenin in Acipenser baerii exposed to 17β-estradiol

摘要 以17β雌二醇(E2)诱导西伯利亚鲟幼鱼,取其血清样品后利用凝胶过滤层析进行分析,诱导组鱼血清中较对照组的多出一蛋白峰;收集浓缩该峰后经离子交换两步层析法进行分离,成功分离出西伯利亚鲟卵黄蛋白原(Vtg)。分离出的Vtg于7.5%的Native-PAGE凝胶电泳分析,得出其分子量为390 KDa;对Vtg氨基酸组成检测结果显示,谷氨酸、丙氨酸、赖氨酸、天冬氨酸和亮氨酸含量较高,分别为12.25%、10.15%、8.43%、8.37%和8.11%,但甲硫氨酸和组氨酸含量相比较低,分别为1.56%和2.42%。 Vitellogenin(Vtg) of Acipenser baerii serum was isolated,analysed and determined after being exposed to 17β-estradiol.Results showed that there was another protein peak comparing to the control group after being exposed to 17β-estradiol in Acipenser baerii serum by gel filtration chromatography.Vtg was effectively purified from the serum of Acipenser baerii by two steps of ion-exchange chromatography.The molecular weight of Vtg was 390 KDa through the Native-PAGE gel electrophoresis.Amino acid composition analysis showed that the Vtg was abundant in glutamic acid(12.25%),alanine(10.15%),lysine(8.43%),aspartic acid(8.37%) and leucine(8.11%),but the content of methionine and histamine was lower,1.56% and 2.42% respectively.
