摘要
目的研究炔雌醇(EE2)对斑马鱼的卵黄蛋白原(VTG)及性腺发育的影响,并通过一系列生殖发育终点指标的研究选择有效筛选环境雌激素(EEs)的敏感指标。方法将孵化后20 d的斑马鱼暴露于10 ng/L EE2,同时设空白对照和溶剂对照。至孵化后60 d时检测各组身长、体重、VTG含量及其基因表达水平。至孵化后120 d,进行性别比例的观察和性腺、肝脏的组织学检查。结果与对照组相比,EE2组斑马鱼的体重、体长降低,P<0.01;VTG含量明显增高,P<0.01;雌鱼比例升高,P<0.01。性腺的光镜检查发现,EE2组的性腺发育滞后于对照组。肝脏电镜检查显示EE2处理组的斑马鱼肝脏内糖原减少。结论EE2能影响斑马鱼的生长发育,诱导鱼体内合成大量VTG,影响性别比例,延缓性腺成熟。VTG的含量及其基因表达水平,身长体重、性别比例、性腺的组织学检查等都可作为监测EEs暴露的有效终点指标,其中VTG可能是最敏感的指标。
Objective To study the effect of ethynylestradiol(EE2 ) on vitellogenin(vtg) and gonad development of zebrafish; to select sensitive biomarkers for screening environmental estrogens using zebrafish. Methods Zebrafish of 20 days post hatch were randomly divided into 10 ng/L EE2 group, blank control group and vehicle control group. At 60 days post hatch, body weight, body length, vtg concentration and its mRNA level of zebrafish were measured. At 120 days post hatch, sex ratio and histological changes in gonad and liver were recorded. Results Compared with control group, body weight and body length of zebrafish decreased ( P 〈 0.01 ) and its vtg concentration increased( P 〈 0.01 ) after EE2 treatment. Histological examination showed that the development of gonad was delayed and hepatin in liver decreased after EE2 treatment. Conclusion EE2 can induce zebrafish to produce large quantity of vtg and have negative effect on its growth. Further more, EE2 may have impact on sex ratio. The indicators, such as concentration of vtg and its mRNA level, body weight and length, sex ratio and histological changes could be the appropriate endpoints for screening environmental estrogens, among which vtg might be the most sensitive biomarker.
