亚硒酸钠对人外周淋巴细胞体外转化与增殖的影响 被引量:15

摘要 目的为了解亚硒酸钠对人周淋巴细胞在体外培养中转化与增殖产生有益或不利影响的临界剂量。方法用不同剂量的亚硒酸钠处理培养中的淋巴细胞72h,观察T细胞转化率及增殖指数。结果在培养物中添加0.05μg/ml亚硒酸钠能显著促进转化细胞增殖;添加0.25μg/ml可促进转化细胞增殖,但显著抑制细胞转化;添加0.75μg/ml和1.50μg/ml,不仅不能促进细胞增殖,而且强烈抑制细胞转化。1.50μg/ml剂量的亚硒酸钠使非转化淋巴细胞发生葡萄状聚集甚至核融合。结论亚硒酸钠影响淋巴细胞增殖的临界剂量在0.25~0.75μg/ml之间,影响其转化的临界剂量在0.05μg/ml以下,淋巴细胞转化较之增殖对亚硒酸钠作用的反应更加敏感。 Objective To understand the critical dose of sodium selenite which shows beneficial or deteriorative effect on the transformation and proliferation of human peripheral blood lymphocytes (PBL) in culture in vitro.Methods Normal human PBL were treated for 72h by supplementing the culture medium with different doses of sodium selenite,and then the transformation rate and proliferation index of the T lymphocytes were studied.Results When the culture medium was supplemented with 0.05μg/ml sodium selenite,the proliferation of transformed cells was significantly promoted and the transformation did not change remarkably,when supplemented with 0.25μg/ml, the proliferation was promoted but the transformation was inhibited,and when with 0.75μg/ml and 1.50μg/ml,the proliferation was not promoted and the transformation was strongly inhibited.Big dose of (1.50μg/ml) sodium selenite made the nuclei of the untransformed lymphocytes gather together like a few grapes doing,and even caused them to fuse.Conclusions The critical dose of sodium selenite which shows beneficial or deteriorative effect on proliferation of human PBL in culture is between 0.25 ̄0.75μg/ml,and the critical dose on transformation below 0.05μg/ml.Lymphocyte transformation is more sensitive than lymphocyte proliferation to the effect of selenium.In addition the toxic concentration of selenium may damage the stability of membrane structure of cells.
