摘要
目的观察二苯乙烯苷对兔血小板内游离钙离子浓度([Ca2+]i)的影响,探讨其抗血小板聚集的作用机制。方法采用比浊法测定兔血小板聚集功能;双波长荧光探针Fura-2测定血小板细胞质内[Ca2+]i。结果 1~10μmol·L-1二苯乙烯苷体外给药对二磷酸腺苷(ADP)和凝血酶引起的血小板聚集有浓度依赖性抑制作用。有无细胞外钙时,1~10μmol·L-1二苯乙烯苷对静息态血小板的[Ca2+]i均无明显影响,但对激活态血小板[Ca2+]i的升高有明显抑制作用,呈剂量依赖性。结论二苯乙烯苷抑制血小板聚集的作用机制可能与其抑制血小板细胞质[Ca2+]i的升高有关。
Objective To investigate the effect of stilbene glucoside on intracellular free calcium concentration of rabbit platelet,and to probe the possible mechanism of its anti-platelet aggregation. Methods Rabbit platelet aggregation and platelet [Ca2+]i were determined by Born method and double beam fluorescence spectrophotometer method,respectively. Results Stilbene glucoside 1-10 μmol·L-1 inhibited the rabbit's platelet aggregation induced by ADP and thrombin in a concentration-dependent manner in vitro. 1-10 μmol·L-1 stilbene glucoside had no effect on the resting [Ca2+]i,but inhibited [Ca2+]i induced by thrombin in a concentration-dependent manner with or without extracellular [Ca2+]. Conclusion Stilbene glucoside markedly inhibits the rabbit platelet aggregation,and the mechanism of which may be related to its depression on the rise of platelet [Ca2+]i.
