青蒿同源四倍体组培优化及青蒿素含量测定 被引量:2

Optimization of Cultivation System and Determination of Arteannuin Content of Autotetraploid Plants of Artemisia annua L.

摘要 采用MS添加不同激素组合的培养基,探讨青蒿同源四倍体快速繁殖的适宜培养基,为最终建立青蒿同源四倍体快繁体系奠定基础。对其田间生长3个月的不同株系进行青蒿素含量测定,旨在筛选出有效成分最高的青蒿同源四倍体株系。结果表明:青蒿同源四倍体丛生芽诱导的最佳条件为MS+6-BA(6-苄氨基嘌呤)0.5 mg/L+IAA(吲哚乙酸)0.1 mg/L;在青蒿6个四倍体株系中,青蒿素含量相对二倍体株系的都有不同程度的提高,青蒿四倍体株系的平均青蒿素含量为二倍体的1.50倍。 To use MS medium with different combinations of hormones to explore the appropriate medium for the rapid propagation system of autotetraploid plants of Artemisia annua L. and eventually test- ablish the foundation for the rapid propagation system of autotetraploid plants of Artemisia annua L. After growing in field for 3 months, HPLC is used to determine Arteannuin content of autotetraploid plants of Artemisia annua L. in order to screen a strain of autotetraploid plants of Artemisia annua L. , which cantains the highest Arteannuin content. The results show that the best condition of rapid propagation for autotetraploid plants is MS+6 - BA 0.5 mg/L+IAA 0. 1 mg/L. The content of artemisinin in autotetraploidy of Artemisia annua L. has increased compared with that in the diploid. Averagely, artemisinin content in the autotetraploid is 1.5 times as much as that in the diploid. Thus it has provided the foundation for breeding and identification of excellent autotetraploid lines of Artemisia annua L.
