摘要
试验从影响原生质体制备的诸多方面进行了研究,探索榆黄蘑原生质体制备的优化系统。试验时取鲜嫩的菌丝,用pH值为60的KCl、蔗糖、甘露醇做酶解稳定液,30℃下水浴酶解,酶解时间为4~5h,此时榆黄蘑原生质体释放量最大,达到107个/mL。
The experiment was researched in many ways which affect protoplast preparation. A superior system of protoplast of Pleurotus citrinopileutus preparation was studied: fresh Fungus threads were hydrolysised by lywallzyme which was dissolved in KCl or annitol or sucrose in different pH conditions.Under the conditions of T=30 and pH =6 and the time of enzyme hydrolysis during 45 h, the burst rate of protoplast was up to max. The rate of releasion isabout 107/mL.
