摘要
目的研究全身γ射线照射后伤口组织生长因子基因表达的变化及苯妥因钠的作用。方法采用置入聚乙烯醇海绵的大鼠背部切口伤模型,用原位杂交技术和计算机图像分析技术测定伤口组织中血小板源性生长因子B(PDGFB)和转化生长因子β1(TGFβ1)mRNA表达的平均吸光度。结果6Gy全身照射后伤口组织中PDGFB和TGFβ1mRNA阳性表达平均吸光度明显下降;苯妥因钠对单纯创伤和全身照射合并创伤的伤口组织中PDGFB和TGFβ1mRNA表达的平均吸光度有明显的提高。结论这说明伤口组织中生长因子基因表达的降低是全身照射后创伤愈合延迟的分子机制之一;苯妥因钠上调伤口组织中生长因子基因表达而有利于创伤愈合。
Objective\ To study the changes of gene expression of growth factors in wound tissues after 60 Co γ irradiation and the role of phenytoin sodium. Methods Wound tissues were collected from dorsum incision in rats,in which polyvinyl alcohol sponges had been implanted.The expressions of platelet derived growth factor B (PDGF B) mRNA and transforming growth factor β 1 (TGF β 1) mRNA in wound tissues were determined by in situ bybridization. Results It was found that the expressions of PDGF B mRNA and TGF β 1 mRNA were decreased after 6 Gy whole body irradiation,and phenytoin sodium significantly increased the expressions of PDGF B mRNA and TGF β 1 mRNA in both irradiated and non irradiated wounds. Conclusion It is suggested that the decrease of expressions of growth factors in wound tissues plays an important role in the impairment of wound healing by systemic irradiation.Phenytoin sodium enhances the expression of growth factors in wound and helps improve wound healing.
