乌鲁木齐汉族儿童哮喘α_1-抗糜蛋白酶表型分析研究 被引量:2

Study of phenotypes of α_1-antichymotrypsin in Hanzu childhood asthma in Urumqi

摘要 【目的】探讨α1-抗糜蛋白酶(α1-antichymotrypsin,α1-ACT)不同表型在乌鲁木齐汉族儿童哮喘发病中的作用。【方法】建立聚丙烯酰胺凝胶等电聚焦免疫固定技术,并应用其对54例哮喘儿童及50例健康儿童α1-ACT蛋白表型进行分析,用统计学方法分析α1-ACT不同表型对儿童哮喘发病的影响。【结果】哮喘组以及儿童对照组α1-ACT均存在I型、II型、III型3种表型,儿童对照组α1-ACTI型表型频率占86%、II型表型频率占10%、III型频率占4%,哮喘组α1-ACTI型表型频率占80%、II型表型频率占6%、III型占22%。α1-ACTI型表型分布在哮喘组和对照组之间差异无统计学意义(χ2=0.638,P>0.05),α1-ACTII型表型分布在哮喘组和对照组之间差异无统计学意义(χ2=0.135,P>0.05);α1-ACTIII型表型分布哮喘组明显高于儿童对照组(χ2=5.659,P<0.05)。【结论】α1-ACTIII型分布增加了乌鲁木齐汉族儿童哮喘发病的危险性。 【Objective】 To investigate whether heterozygous deficiency of α1-antichymotrpsin(α1-ACT) is a factor of the pathogenesis of childhood asthma in Urumqi. 【Methods】 50 children with asthma and 50 healthy children's phenotypes of α1-ACT in the Han Population in Urumqi had been determined by isoelectric focusing on the polyacrylamide gel followed by immunofixation.The phenotype frequencies of α1-ACT were investigated using statistical method and were examined for serum level of α1-ACT with rocket immunoelectrophoresis. 【Results】 Three phenotypes of α1-ACT were found in asthmatic group and control group.The rate of phenotype I of α1-ACT was 86% and phenotype II of α1-ACT was 10% while phenotype III of α1-ACT was 4% in control group.The rate of phenotype I of α1-ACT was 80% and phenotype II of α1-ACT was 6% while phenotype III of α1-ACT was 22% in asthmatic group.Distribution of phenotype I of α1-ACT had no significantly diffirence between two groups(χ2=0.638,P0.05).Neither had distribution of phenotype II of α1-ACT no significantly diffirence between two groups(χ2=0.135,P0.05).Rate of phenotype III of α1-ACT was higher in asthmatic group was lower than that in control group(χ2=5.659,P0.05). 【Conclusion】 This study provides the evidence that phenotype III increases the risk of developing asthma during childhood in Urumqi.
