摘要
目的:通过研究细胞因子和泌乳素(PRL)对球后成纤维细胞(RFs)表面细胞间粘附分子-1(ICAM-1)表达的影响,进一步了解甲状腺相关眼病(TAO)的发病机理,以探索新的防治途径。方法:将体外培养的正常人RFs与IFN-γ、IL-4、IFN-α和PRL孵育72h,利用直接免疫荧光技术和流式细胞仪检测其表面ICAM-1表达的情况。结果:正常人RFs中只有极少数细胞自发并较弱地表达ICAM-1,基础状态下的阳性百分数为(3.49±1.99),平均荧光强度为(44.84±20.59);而IFN-γ、IL-4、IFN-α能以剂量依赖方式显著增强ICAM-1的表达(P<0.05)。PRL不仅可以直接刺激RFs表面ICAM-1的表达(P<0.05),其剂量相关曲线呈双相型,还能拮抗或协同IFN-γ(100u/ml)、IFN-α(1000u/ml)或IL-4(2.5ng/ml)的诱导作用。结论:IFN-γ、IL-4、IF-α和PRL对正常人RFs表面ICAM-1的表达有明显的调节作用,它们可能是启动或加重TAO自身免疫反应的重要因素。
Objective: To further understand the pathogenesis of thyroid associated ophthalmopathy and develop new curative ways, we studied the influences of cytokines and prolactin (PRL) on ICAM-1 expression on the surfaces of retroocular fibroblasts (RFs). Methods: With direct immunofluorescence technique and flowcytometry, we determined ICAM-1 expression on the surfaces of cultured normal human RFs incubated with IFN γ,IL 4,IFN α and(or) PRL for 72 hours. Results: Very few RFs spontaneously and weakly expressed ICAM 1, IFN γ,IL 4 and IFN α significantly enhanced the expression of ICAM 1 in dose-dependent manners ( P <0 05). In addition, PRL could not only directly stimulate ICAM 1 expression on the surfacts of RFs ( P <0 05), which dose-dependent curve showed “inverted U shape”, but also antagonize the effects of IFN γ (100 u/ml) as well as IFN α (1000 u/ml) and coact with IL 4 (2.5 ng/ml). Conclusion: IFN γ, IL 4, IFN α and PRL played significant roles in regulating ICAM 1 expression on the surfaces of normal human RFs and they might be the important factors for initiating or promoting autoimmune reactions in thyroid associated ophthalmopathy.
