摘要
目的在基因水平上定量分析并探讨人载脂蛋白C-Ⅲ(apolipoproteinC-Ⅲ,apoC-Ⅲ)基因多态性与颈动脉粥样硬化(CAS)的相关性。方法选择住院患者和体检者共700例,根据彩色多普勒超声检查是否有CAS形成,分为CAS组(400例)和对照组(300例)。取静脉血行血生化检测和提取全血DNA,结合Taqman探针,通过荧光实时定量PCR,同时分析apoC-Ⅲ—625和—455位点变异与CAS、血脂的关系。结果 CAS组患者apoCⅢ—455C纯合子基因型和—625del—455c基因型的比例明显高于对照组(70.7% vs 52.8%,50.0% vs 25.2%,P<0.05)。CAS伴高TG、单纯高TG受试者apoC-Ⅲ—455C基因的量明显低于CAS不伴高TG和非CAS非高TG受试者(P<0.01)。apoC-Ⅲ—455C基因的量与CAS风险呈正相关。结论 apoC-Ⅲ—455C基因通过脂代谢异常增加CAS的风险。—455C纯合子基因型携带者具有高的患CAS风险。apoC-Ⅲ-625del与-455C有协同增加CAS风险的作用。
Objective To explore the relation between apolipoproteinCⅢ（apoCⅢ） gene polymorphisms and carotid atherosclerosis（CAS）.Methods Peripheral blood samples were collected from 400 CAS patients and 300 people without CAS.Genomic DNA was extracted from whole blood samples.The relation between variants of apoCⅢ-625 and -455 and CAS was studied by fluorescence quantitative PCR.The relation between variants of apoCⅢ-625 and -455 and the concentration of lipid was analyzed.Total cholesterol,triglyceride（TG）,highdensity lipoprotein cholesterol,lowdensity lipoprotein cholesterol and apoCⅢ were measured.Results The apoCⅢ-455C and -625del-455C homozygote rates of the CAS patients were significantly higher than those of control group（70.7% vs 52.8%,50.0% vs 25.2%,P〈0.05）.The apoCⅢ-455C expression rates of the patients with high TG and high TG accompanied by CAS were lower than those of the patients with CAS and without high TG and the patients without CAS and high TG.The quantity of apoCⅢ-455C was positively correlated to the risk of CAS.Conclusion The gene polymorphisms of apoCⅢ-455C increase the risk of CAS by disturbing lipid metabolism.The patients with homozygotes of the apoCⅢ-455C have high risk of CAS.apoCⅢ-455C cooperating with -625del will increase the risk of CAS.
