摘要
目的观察益气养阴活血方对糖尿病肾病大鼠肾皮质糖基化终产物(AGEs)及其受体(RAGE)mRNA表达的影响,探讨其治疗糖尿病肾病的作用机制。方法用链脲佐菌素腹腔注射制备糖尿病肾病大鼠模型,模型大鼠随机分为正常组、模型组、贝那普利组、益气养阴活血方组(简称中药组),并用相应的药物干预,给药8周后检测各组大鼠肾皮质中AGEs的含量,用免疫组化法检测肾皮质中AGEs的蛋白表达,用RT-PCR的方法检测肾皮质RAGEmRNA的表达。结果糖尿病肾病大鼠肾皮质AGEs含量明显增加,RAGEmRNA出现过度表达。经中药治疗8周后肾皮质AGEs含量明显减少,由(513.26±97.2)AUF/mg降为(417.35±89.9)AUF/mg;RAGEmRNA表达明显下调,由(1.51±0.01)降为(0.62±0.04),P<0.01。结论益气养阴活血方能够减少AGEs在肾皮质的积累,下调糖尿病肾病大鼠肾皮质RAGEmRNA的过度表达。
Objective To observe the influences of the formulas with the actions of replenishing qi,nourishing yin and activating blood circulation on the expressions of advanced glyclation end-products(AGEs) and mRNA of receptor of advanced glyclation end-products(RAGE-mRNA) in rats with diabetic nephropathy(DN) ,and to discuss the mechanism of the formulas in the treatment of DN. Methods The rat model of DN was established by the intraperitoneal injection of strepotozotocin(STZ) .All rats were randomly divided into the normal group,model group,Benazepril group and Chinese medicine group and all groups were given relevant medicinals respectively.After 8 weeks the content of AGEs were detected in the renal cortex in DN rats.The expression of AGEs was detected by using immunohistochemistry assay,and the expression of RAGE-mRNA was detected by using RT-PCR. Results The content of AGEs in the renal cortex increased significantly and RAGE-mRNA expressed excessively in DN rats.After treatment with the formulas for 8 weeks,the content of AGEs decreased significantly,from(513.26±97.2) AUF/mg to(417.35±89.9) AUF/mg,and the expression ofRAGE-mRNA decreased significantly,from(1.51±0.01) to(0.62±0.04) ,P0.01.Conclusion The formulas with the actions of replenishing qi,nourishing yin and activating blood circulation can reduce the accumulation of AGEs in the renal cortex,and regulate downwards the over-expression of RAGE-mRNA in DN rats.
