摘要
目的:探讨中药复方糖肾康胶囊对早期糖尿病肾病的肾脏保护机制。方法:大鼠随机分为对照组(A组)、模型组(B组)、糖脉康治疗组(C组)、伊那普里治疗组(D组)、糖肾康治疗组(E组),每组12只。观察56d后,用RT-PCR检测各组肾皮质TGF-β_1mRNA的表达。检测体重、血溏、血尿素氮(BUN)、血肌酐(Cr)、尿白蛋白排泄率(UAER)、肾重指数。结果:B组56d后出现体重显著降低(P<0.01);血糖、肾重指数、血BUN与Cr水平显著升高(P<0.01);肾皮质TGF-β_1mRNA的表达明显增高(P<0.01)。经过糖肾康56d的治疗后,体重显著回升(P<0.01);血糖、肾重指数、UAER显著降低(P<0.01);肾皮质TGF-β_1mRNA的表达明显降低(P<0.01)。结论:糖肾康对早期糖尿病肾病的肾脏具有保护作用。其机制可能为糖肾康下调TGF-β系统的表达,后者有抑制细胞肥大、减少细胞外基质成分的合成的作用。
Objective: To explore the protection of Chinese medicine prescription of tangshenkang capsules on diabetic nephropathy. Methods: Rats were devided into control group (A group), model group (B group), tangmaikang group (C group), Yina Pull group (D group), tangshenkang group (E group), and each group wad ten rats. After treated eight weeks, the expression of TGF -β 1 mRNA in cortex of kidney was detected by RT - PCR. The weight, blood glucose, blood urea nitrogen (BUN), serum creatinine (Cr), urinary albumin excretion rate, index of kidney weight were also detected. Results. After eight weeks, weight of rats in B group was remarkably lowered (P 〈 0.01 ), and the level of blood glucose, index of kidney weight, BUN, Cr were remarkably risen (P〈0.01), and the expression of TGF- β1 mRNA in cortex of kidney was remarkably risen (P 〈 0.01 ). After treated eight weeks, weight of rats was rernarkably risen (P 〈 0.01 ), and the level of blood glucose, index of kidney weight, UAER were remarkably lowered (P〈 0.01), and the expression of TGF-β1 mRNA in cortex of kidney was remarkably lowered ( P 〈 0.01). Conclusion: tangshenkang capsules could protected the kidney of early diabetic nephropathy, and it's mechanism was tangshenkang could lower the expression of TGF- β. TGF-β could inhibit cells multiplication and reduce the synthesis of extracellular matrix.
