口服咖啡因对早期妊娠小鼠胚泡植入的影响

摘要 目的研究咖啡因对小鼠妊娠早期胚泡植入的影响。方法将孕鼠随机分为4组(每组20只):低、中、高剂量咖啡因组(分别给予60、120、240mg/kg剂量的咖啡因)和对照组(给予等量生理盐水)。观察口服不同剂量咖啡因的妊娠小鼠胚泡着床的变化,并进一步分析雌激素和孕酮的变化,以及着床点部位子宫内膜厚度、腺体发育和糖原含量的变化。结果高剂量咖啡因组[240mg/(kg·d)]孕鼠着床率显著低于其他两个试验组和对照组(P<0.05),同时发现低剂量组[60mg/(kg·d)]和中剂量组[120mg/(kg·d)]雌激素水平显著高于对照组和高剂量组(P<0.05);中剂量组孕酮水平显著高于其他三组(P<0.05)。低、中浓度咖啡因处理组着床点部位子宫内膜厚度比对照组显著增加,而高浓度咖啡因处理组着床点部位子宫内膜厚度则显著低于其他三组(P<0.05)。与对照组和低、中浓度咖啡因处理组相比,高浓度咖啡因处理组子宫内膜腺腔直径缩小,腺体数量显著减少;另外还观察到着床点部位子宫内膜糖原含量的变化与血清激素水平的变化是一致的。结论口服高剂量咖啡因可引起雌、孕激素分泌紊乱,阻滞子宫内膜的蜕膜化反应,进而影响早期妊娠小鼠的胚泡植入。 Objective To investigate the effect of oral caffeine on blastocyst implantation to early pregnant mice. Methods Eighty pregnant mice were randomly divided into 4 groups (20 each):low caffeine dose group [60mg/(kg·d)],medium caffeine dose group [120mg/(kg·d)],high caffeine dose group [240mg/(kg·d)] and control group (same volume of normal saline). Changes in embryo implantation in pregnant mice treated with different doses of caffeine were observed. At the same time,observations and analyses were done for the levels of serum estrogen and progesterone,the changes in endometrial thickness,endometrial glands,and endometrial glycogen contents. Results The successful implantation rate was significantly lower in high caffeine dose group than in the other two dose groups and control group. The level of serum estrogen was remarkably enhanced in both medium and low caffeine dose groups,and the level of serum progesterone was significantly higher in medium caffeine dose group than in the other groups. It was found that the changes in endometrial glycogen were consistent with the changes in serum hormones. Conclusion Oral administration of high dosage of caffeine may lead to the parasecretion of oestrogen and progestogen,block the decidualization of endometrium,and accordingly affect the blastocyst implantation in early pregnant mice.
