摘要
目的白血病细胞能够通过自分泌方式分泌细胞因子而作用于自身,使细胞大量的增殖;白血病细胞所分泌的细胞因子也能作用于人脐带血(UCB)单个核细胞(MNCs)中的CD34+/CD133+细胞亚群使之增殖。关于HL-60细胞(急性早幼粒白血病细胞)培养上清液能否使UCB-MNCs增殖尚未见报道,本实验主要目的是观察HL-60细胞培养上清液对人UCB-MNCs增殖的影响。方法实验分为①有HL-60细胞培养上清液+StemSpanSFEM组;②StemSpan誖SFEM组;③HL-60细胞培养上清液+高糖(HG)-DMEM组;④HG-DMEM组;⑤HL-60细胞培养上清液+低糖(LG)-DMEM组;⑥LG-DMEM组。收分离的人UCB-MNCs按2.5×106/瓶分别接种于6组培养液中,37℃,5%CO2,饱和湿度培养,分别于第3,第6,第9,第12天计细胞总数。结果①培养至第3天,HL-60细胞培养上清液+StemSpanSFEM组和StemSpanSFEM组MNCs数较培养前和其余各组明显升高(均P<0.05),至第9天细胞数维持在较高水平;②HL-60细胞培养上清液+StemSpanSFEM组和StemSpanSFEM组各时点上,前者MNCs数量较后者有所增加,但无统计学意义(P>0.05);③添加HL-60上清的高糖组和低糖组,各时点上的MNCs数均较相应的对照组升高(均P<0.05)。结论 HL-60细胞培养上清液对人UCB-MNCs具有一定的增殖作用,其有效生物活性物质有待进一步研究。
Objective Leukemic cells can excrete cytokines by an autocrine way,which contribute to its large-scale proliferation,and the cytokines may also contribute to the proliferations of CD34+/CD133+ subpopulation of UCB-MNCs too.To the effect of HL-60 cell supernatant on the proliferation of UCB-MNCs,there is not related report so far.The aim of this study is to investigate effects of HL-60 cell supernatant on proliferations of mononuclear cells from human umbilical cord blood.Methods Six various cultural system are listed as follows: ① HL-60 cell supernatant+ StemSpanSFEM;② StemSpanSFEM;③ HL-60 cell supernatant+ HG-DMEM;④ HG-DMEM;⑤ HL-60 cell supernatant LG-DMEM;⑥ LG-DMEM.2.50×106 of UCB-MNCs per bottle with the above each cultural system is incubated at 37℃ in a 5%CO2,saturation humidity of incubator,the number of cell each group calculated by trypan blue assay on 3rd,6th,9th,and 12th day.Results ① On 3rd day,the numbers of MNCs,both HL-60 cell supernatant+StemSpanSFEM and StemSpanSFEM groups are increased significantly than other groups’(all P0.05),and kept a steady cell number.② As compared to HL-60 cell supernatant+StemSpanSFEM and StemSpanSFEM groups,the number of MNCs of former is higher at any time,but no statistically significant(P0.05);③ Number of MNCs,both in HG-DMEM and LG-DMEM medium with HL-60 cell supernatant,are higher than control group without HL-60 cell supernatant(all P 0.05).Conclusions HL-60 cell supernatants may contribute to the proliferation of UCB-MNCs,and its related bioactive compound need further investigation in the near future..
