摘要
目的:验证温阳解郁方对抑郁症的治疗作用并探讨其作用机理。方法:以出生后经过10d母婴分离的成年C57BL/6J小鼠遭受社会击败应激塑造抑郁症模型,用温阳解郁复方温阳解郁方进行为期4周的治疗性干预,通过旷场、高架十字迷宫及决择反应等实验观察行为模式变化,运用放射免疫法检测应激后和DEX/CRH实验检测血清皮质酮的变化,运用免疫组织化学方法检测中枢糖皮质激素受体(GR)及11β-羟基类固醇脱氢酶2(11β-HSD2)的表达。结果:温阳解郁方可明显改善母婴分离社会击败应激模型小鼠抑郁焦虑行为和促进趋向决择反应,可在一定程度上改善下丘脑-垂体-肾上腺(HPA)轴功能。结论:温阳解郁方对抑郁症有明显治疗作用。
Objective：Further verify therapeutic effect of Warming Yang with Soothing Liver Formula（Xie Shen Fang,XSF） on depress,to explore its mechanism.Methods：The adult C57BL/6J mice which experience 10d maternal separation after birth,were subjected 10d social defeat stress to make depression model,then treated with XSF for 4w,observed behavior changes by open field,elevated plus maze and decision-making test,detected serum corticosterone by radioimmunoassay,central glucocorticoid receptor（GR） and 11β-hydroxysteroid dehydrogenase 2（11β-HSD2） expression by immunohistochemistry.Results：（1） XSF significantly regulated anxiety tend of depression mice and promote appetitive decision-making response;（2） XSF improved hypothalamus-pituitary-adrenal（HPA） axis function at certain extent.Conclusion：Warming Yang with Soothing Liver Formula show clear therapeutic effect on depression mice and would be better to prolong treatment course depression mice and would be better to prolong treatment course.
