In this article,we systematically introduced the basic principles and protocols of four methods,co-transformation,transposable element,site-specific recombination, and intrachromosomal recombination, respectively, applied in selectable marker gene knockout in transgenic plants. The prospect of technology of selectable marker gene knockout was offered, through the analysis of their respective features as well as practical applications and the comparison of their advantages and disadvantages. The development of selectable marker gene knockout plays an active role in not only the security and commercialization of transgenic crops, but also the development and improvement of transgenic technology.
