葎草乙酸乙酯提取物抗炎镇痛作用及其机制的研究 被引量:14

摘要 目的:研究葎草乙酸乙酯提取物(LAT)的抗炎镇痛药理作用,并对其作用机制进行初步研究。方法:采用醋酸扭体法实验和热板法实验两种疼痛模型观察葎草的镇痛作用;采用二甲苯致小鼠耳肿胀、棉球诱导大鼠肉芽组织增生和鸡蛋清致大鼠足肿胀法实验模型研究葎草的抗炎作用;通过小鼠福尔马林镇痛实验和测定大鼠足爪炎性渗出物中PGE2含量研究其抗炎镇痛机制。结果:葎草乙酸乙酯提取物能明显的减少小鼠的扭体次数,显著的提高小鼠的痛阈值,对二甲苯致小鼠耳肿胀、棉球诱导大鼠肉芽组织增生和鸡蛋清致大鼠足肿胀均有明显的抑制作用;并能够降低小鼠福尔马林镇痛实验中Ⅱ相疼痛反应,能降低大鼠足爪炎性渗出物中PGE2的含量。结论:葎草醇乙酸乙酯提物具有明显的抗炎镇痛作用,葎草醇乙酸乙酯提物具有一定的外周性镇痛活性,其抗炎作用可能与其抑制炎性组织中的PGE2合成或释放有关。 Objective:To study on the anti-inflammatory and analgesic activities of ethyl acetate extract from Humulus scandens (Lour.)Merr.and its underlying mechanism.Methods:Acetic acid writhing method and hot-plate method were used to study the analgesic activities.Auricle edema induced by dimethylbenzene,cotton pellet granuloma of the mice and edema in feet induced by egg whites were applied for anti-inflammatory experiments.The underlying mechanism was performed by formaldehyde method and determination on the content of PGE2 in inflammatory tissue.Result:The ethyl acetate extract from Humulus scandens (Lour.)Merr.inhibited acid-induced mouse writhing response and enhanced the threshold of pain value significantly.The ethyl acetate extract inhibited auricle edema induced by dimethylbenzene and edema in feet induced by egg whites significantly.The ethyl acetate extract significantly inhibited the size of cotton pellet induced granuloma formation.The ethyl acetate extract had inhibitory effect onⅡphase pain in formalin inducing pain.The extract could reduce the content PGE2 in inflammatory tissue.Conclusion:The ethyl acetate extract from Humulus scandens (Lour.)Merr.shows anti-inflammatory and analgesic activities.The extract showed analgesic activity through inhibition subsequent analgesic reaction.The anti-inflammatory activities may relate to its inhibitory effects on the content of PGE2 in inflammatory tissue.
