胰岛β细胞株INS-1和MIN6有关糖毒性的比较研究 被引量:3

摘要 目的探讨INS-1和MIN6细胞株在有关糖毒性研究的不同表型。方法胰岛素瘤细胞株INS-1和MIN-6分别在含5和25mM糖浓度的无血浆培养基中孵育24h。MTT法测定细胞活性,细胞凋亡通过annexinV-PI双染,流式细胞仪检测分析。结果 INS-1细胞在高糖条件下细胞活性明显降低,细胞凋亡率、死亡率明显增加,而MIN6细胞在高糖环境下细胞活性增加,凋亡率下降,死亡率无明显变化。结论 MIN6细胞比INS-1细胞能耐受更高的糖毒性。 [Objective] To investigate different glucotoxicity-associated phenotypes in pancreatic β-ceil line INS-1 and MIN6. [Methods] After incubation for 24h in serum-free media containing 5mM or 25raM glucose respectively, INS-1 and MIN-6 cells were subjected to cell viability detection by MTT method, and apoptosis analysis was made by annexinV-PI apoptotie kit detection and flow cytometry. [ Results ] Under condition of 25 mM glucose, INS-1 cells displayed signitieant decrease in cell viability and increase in apoptosis and necrosis, whereas MIN6 cells exhibited noticeably increased cell viability, decreased apoptosis and inapparently necrosis. [Conclusion] Comparing with INS-1 cells, MIN6 ceils are more tolerable to glucotoxicity.
