摘要
目的:观察补肺汤对博莱霉素致肺纤维化大鼠肺组织中转化生长因子(TGF-β1)表达的影响。方法:SD大鼠96只,随机分成正常组、模型组、补肺汤治疗组和强的松组,每组24只。除正常对照组给予生理盐水外,其余各组均予气管内注射博莱霉素诱导肺纤维化。各组在造模后第2天分别予生理盐水、生理盐水、补肺汤流浸膏、强的松混悬液灌胃,并于造模后第7、14、28天分批分组处死8只大鼠。取右肺行HE染色,观察肺组织形态学变化,免疫组化观察各组大鼠肺组织中TGF-β1蛋白的表达。结果:与模型组比较,补肺汤组和强的松组的病理形态学变化明显减轻,肺泡炎及纤维化积分明显降低;肺组织TGF-β1蛋白表达明显降低(P<0.01)。结论:抑制肺组织TGF-β1的表达可能是补肺汤抗肺纤维化作用的机制之一。
Objective:To observe the effect of bufei decoction on TGF-β1 expression in lung tissue of rats with pulmonary fibrosis induced by bleomycin.Methods:96rats were divided into four groups:normal,model,positive and treated groups,each group contained 24 rats;each group was given bleomycin intratracheal injection to build the pulmonary fibrosis models except normal group,which was given saline.Each group was given the corresponding medicine on the second day after modeling,normal and model groups were given saline,positive group was given prednisone,treated group was given bufei decoction;8rats of each group werere spectively killed after modeling 7,14 and 28 days,the TGF-β1 expression in lung tissue of rats was determined by immunohistochemical method.Results:The TGF-β1 expression in lung tissue of normal rats was obviously lower than that of the model group,to compare with that of the model group rats,the TGF-β1 expression in lung tissue of rats in prednisone and bufei decoction groups was significantly lowered.Conclusion:Bufei decoction could inhibit pulmonary fibrosis by controlling the expression of TGF-β1 expression in lung tissue.
