摘要
目的探讨三氧化二砷(As2O3)对人胶质瘤U251细胞的生长及端粒酶活性的影响。方法采用倒置显微镜和透射电镜观察As2O3处理后U251细胞形态变化;四甲基偶氮唑蓝比色法观察As2O3对U251细胞增殖的影响;流式细胞术检测细胞凋亡;端粒重序列扩增酶联免疫吸附实验(TRAP-ELISA)结合聚丙烯酰胺凝胶电泳(TRAP-PAGE)银染法检测As2O3处理后U251细胞端粒酶活性变化。结果倒置显微镜下观察到:As2O3处理后的U251细胞逐渐变圆、脱壁,细胞间接触变松,细胞质中颗粒增多,增殖变慢,细胞周围碎片增多;透射电镜下见较多典型凋亡细胞。1～8μmol/LAs2O3明显抑制U251细胞增殖,诱导其凋亡;并使端粒酶活性逐渐下降,该作用呈浓度和时间依赖性。结论 As2O3对人胶质瘤U251细胞株生长具有显著抑制作用,其机制可能与As2O3能够抑制U251细胞的端粒酶活性密切相关。
Objective To investigate the influence of arsenic trioxide（As2O3） on the growth and telomerase activity of human glioblastoma U25l cell line.Methods Inverted microscope and transmission electron microscope was used to visualize apoptotic changes of U251 cells after treatment of As2O3.The methyl thiazolyl tetrazolium assay was used to assess cell proliferation.Cell apoptosis was analyzed by flow cytometry and telomerase activity of the U251 cells was measured by telomeric repeat amplification protocol-ELISA（TRAP-ELISA） and TRAP-polyacrylamide gel electrophoresis（TRAP-PAGE） silver-staining.Results Inverted microscopy showed that the As2O3-treated cells became round and detached,the contact between cells became loose,particles in the cytoplasm and debris around cell increased,and proliferation became slow.Transmission electron microscope showed many classic apoptotic cells.As2O3（1-8 μmol/L） inhibited the proliferation of U25l cells markedly,induced apoptosis,and reduced telomerase activity in a time-and dose-dependent manner.Conclusions As2O3 can inhibit the growth of human glioblastoma U25l cell line,which may be closely related to inhibitory effect of As2O3 on the telomerase activity of U25l cells.
