细胞因子参与下的脐血造血干细胞体外扩增

Amplification of umbilical cord blood haemopoietic stem cell in vitro cultured with human recombinant cytokine

摘要 目的观察细胞因子参与下脐血干细胞体外扩增效果。方法用含15%胎牛血清改良细胞培养液,加入不同的细胞因子,设置1个对照组和3个实验组,分别于培养第0d、3d、7d、14d取培养液,台盼蓝染色计数单个核细胞的总数、流式细胞仪分析CD34+细胞含量。结果与对照组相比,实验组单个核细胞的数量和CD34+细胞的百分比均有显著增加,尤其以Ⅲ组(IL-3,6,2,4)第7d的扩增为最高:单个核细胞总数达(17.11±6.12)×109个/L,CD34+细胞比率达(9.92±2.56)%。结论细胞因子可以提高脐血单个核细胞和CD34+细胞的扩增效率,SCF+IL-3+6+2+4的组合在第7d时达到了最佳的扩增。 Aim To observe the results of amplification of umbilical cord blood haemopoietic stem cell in vitro cultured with different human recombinant cytokine(HRC).Methods The mononuclear cells separated from the collected cord blood were cultured in vitro with modified Dulbecoo's medium(IMDM).Three experimental groups were set up according to the involvement of HRC.Mononuclear cells were counted by trypan blue exclusion and the CD34+ cells were sorted by flow cytometry on the 3rd,7th,and 14th day after culture.Results Compared to the control group,the amount of mononuclear cells and the percentage of CD34+ cells in each experimental group increased significantly.The quantity of mononuclear cells(17.11±6.12)×109 个/L and the percentage of CD34+ cells(9.92±2.56)% in group Ⅲ were the highest on the 7th of culture.Conclusion The efficiency of amplifiaction of cord blood mononuclear cells and CD34+ cells can be enhanced with addition of cytokine.Optimum ampplification was achieved by combination of SCF+IL-3+6+2+4 on 7th day of culture.
