摘要
目的:研究FHIT蛋白在非小细胞肺癌(NSCLC)中的表达及其与Survivin和Ezrin蛋白表达的关系,探讨其在NSCLC病变中的作用及其临床意义。方法:应用组织芯片技术结合免疫组化法检测60例NSCLC组织、54例肺良性病变组织中FHIT、Survivin和Ezrin蛋白的表达。结果:FHIT蛋白在NSCLC中的阳性表达率为51.7%(31/60),显著低于肺良性病变的87.0%(47/54)(P<0.01);FHIT的表达与性别、淋巴结转移及组织学类型相关(P<0.05),与患者的年龄、肿瘤大小、临床分期、肿瘤细胞的分化程度无关(P>0.05);NSCLC组织中FHIT表达与Survivin和Ezrin表达呈显著负相关(r=-0.636,P<0.01;r=-0.714,P<0.01),而Survivin和Ezrin表达无相关关系(r=0.157,P>0.05)。结论:NSCLC中FHIT失表达,Survivin和Ezrin高表达,FHIT、Survivin和Ezrin在NSCLC的发生、发展中相互作用,联合检测FHIT、Survivin和Ezrin的表达有助于判断肺癌的预后。
Objective:To detect the expression of FHIT and the relationship with expressions of Survivin and Ezrin proteins in non-small cell lung cancer(NSCLC),and expore their clinicopathological significance.Methods:The expressions of FHIT,Survivin and Ezrin proteins were detected in 60 samples of NSCLC and 54 specimens with benign lung disease by a tissue microarray and immunohistochemical assay.Results:The positive expression rate of FHIT protein was 51.7% (31/60) in NSCLC tissues,which was lower than that of benign lung disease tissues of 87.0%(47/54)(P0.01).FHIT expression was associated with sex,lymph node metastasis and histological types(P0.05),but not with age,tumor size,clinical staging and level of cellular differentiation(P0.05).FHIT expression was negatively related with Survivin and Ezrin expression in NSCLC tissues(r=-0.636,P0.01;r=-0.714,P0.01),but Survivin expression was not correlated with Ezrin expression(r=0.157,P0.05).Conclusion:Loss of FHIT expression and high expression of Survivin and Ezrin are found in NSCLC.FHIT,Survivin and Ezrin play a synergistic role in the genesis and progression of the malignancy.Combined detection of FHIT,Survivin and Ezrin expression can help in evaluating the prognosis of NSCLC.
