摘要
目的:观察和分析细胞程序性死亡(PCD)在细胞凝聚区形成中的作用和意义。方法:建立和制备C57BL/6N系小鼠胚胎期13、14、15、16d和生后10d等多时间点的颌面部发育模型,采用原位末端转移酶标记法标记颌面部组织的PCD细胞。结果:在颌面部骨、牙齿等多种组织器官形成初期都包括间充质细胞的聚集,并出现特定的细胞凝聚区。随着细胞凝聚的出现,PCD的数量和密度增多。当凝聚区内的细胞向特定细胞系表型转化时,PCD数量下降。结论:PCD在颌面部凝聚区的形成中发挥着重要的作用。
Objective: To observe and analyse the role and effects of programmed cell death (PCD) in the formation of cellular condensation in maxillofacial development. Method: The model of maxillofacial developent of C57BL/6N mouse was established and the series samples of 13, 14, 15 and 16 days of gestation and that of 10 days after birth were processed. PCD was labelled by using TdT mediated dUTP nick end labeling (TUNEL). Results: It was observed that condensation was involved in the development of bone, tooth and other tissues in maxillofacial region. The amount of PCD-positive cells increased after the cell aggregation initialed, in contrast, the number of PCD positive cell decreased when the cells in condensation were undergoing specialized differentiation. Conclusion: PCD is a critical factor in the formation of cellular condensation and PCD contributes in both controlling of the quality and quantity of cells in the condensation and the remoudling of the condensation.
