HL-60细胞在诱导分化中DNA含量及细胞动力学观察

摘要 以显微分光光度法和流式细胞计观察经维甲酸(RA)或二甲亚砜(DMSO)处理 HL-60细胞的 DNA含量和细胞增殖的动态变化.结果显示 HL-60细胞的 DNA 含量由原来高出一倍以上降至接近正常.其中 RA处理细胞 DNA 在处理后第3天和第5天有二次剧降,而 DMSO 处理细胞 DNA 量呈平缓逐日下降.两种处理细胞在诱导第2天 G_1/G_0期细胞明显增多,并在第3天达到高峰。对两种诱导剂作用机理进行了讨论. The alterations of DNA level and cell proliferation dynamics in HL-60 cells induced by RA or DMSO were observed.The results using MSP and FCM showed that DNA level decreased on the 3rd and 5th day in RA treated calls,but in cells treated with DMSO the DNA level gradually decreased day by day.The G_1/G_0 ratio increased markedly on 2nd day attained the peak on 3rd day both RA and DMSO treatment cells.The mechanism of iuduction of differentiation of HL-60 cells by RA and DMSO was discussed.
