摘要
[目的]获得杏鲍菇高产菌株。[方法]通过紫外诱变、单菌落分离、突变株选择、菌丝生长及出菇对比试验研究了杏鲍菇高产菌株的诱变育种技术。[结果]将经紫外线照射处理的孢子悬液稀释,涂在平板培养基上,7 d后分离单菌落并两两配对。取镜检有锁状联合的菌株菌落与原始菌株做拮抗试验,得到了11株突变株。7号和11号菌株与其他菌株间有极显著差异,5号和10号菌株的生长速度显著高于原始菌株,2号菌株与对照菌株相比存在显著差异,其余菌株与原始菌株之间无显著差异。9个菌株的产量存在显著差异,其中5号和10号菌株的产量极显著高于原始菌株,分别提高了35%和34%。它们被命名为PL-5和PL-10。[结论]经紫外线诱变选育的杏鲍菇菌株表现出良好的高产性能。
[Objective] The aim was to obtain high yield Pleurotus eryngii strains.[Method] The mutation breeding technology of high yield P.eryngii strains was studied through UV mutation test,isolating mono-colony,selecting mutate,mycelial growth and comparative mushroom-production test.[Result] The spore suspension treated with UV irradiation was diluted and smeared on plate medium.The mono-colonies were isolated after 7 d for pair-matching.The colonies of strains with clamp connection in microscopic detection were used for antagonism test of initial strains,then 11 mutates were obtained.Strains No.7 and No.11 had extremely significant difference from the other strains.The growth speeds of strains No.5 and No.10 were significantly higher than that of initial strains.Strain No.2 had significant difference from control strains and the other strains had no significant difference from initial strains.The yields of the 9 strains had significant difference,the yields of strains No.5 and No.10 were extremely significantly higher than that of initial strains among them and their yields were increased by 35% and 34% resp.They were named PL-5 and PL-10.[Conclusion] The P.eryngii strains were bred by UV mutation showed good high yield performance.
