摘要
建立微量接种菌落计数法测定体外抗生素后效应。方法:采用微量接种菌落计数法和经典的菌落计数法同时描记金黄色葡萄球菌ATCC25923的生长动力曲线;两种方法同时测定环丙沙星、庆大霉素对ATCC25922、ATCC25923的抗生素后效应。结果:两种方法测定抗生素后效应的结果日内、日间差异无显著性(P>0.05)。结论:微量接种菌落计数法测定抗生素后效应较经典的菌落计数法操作简单、快速,且可节省人力、物力。
OBJECTIVE:A new method for in vitro post antibiotic effect(PAE) measurement was established,being termed micro inocualtion colony counting.METHODS:Growth kinetics curve of S.aureus ATCC 25923 was studied using micro inoculation colony counting and conventional viable counting.PAE for E.coli ATCC 25922 and S.aureus ATCC 25923 were determined by the two methods after 1 h of exposure to ciprofloxacin or gentamicin at a concentration four times the MIC.RESULTS:The results indicated that the mean PAE measurements determined by both methods were very similar and statistically not significantly different( P > 0.05 ). There was little within day and between day variability found among the PAEs for both methods( P > 0.05 ).CONCLUSIONS:The micro inoculation colony counting is considerably less laborious,less time and material consuming than conventional method.
